Description/Mechanism

Specific mechanism is unknown. It is a sedative hypnotic used in the induction and maintenance of anesthesia. Rapid onset and minimal excitation during induction.

Induction/maintenance of anesthesia

Conscious sedation

Contraindications

Hypersensitivity to propafol or propafol emulsion which contains sybean oil and egg phoshpatide.

Obstetrical procedures

Increased intracranial pressure

Adverse Reactions

Twitching or jerking

Clonic movements

Hypotension

Asystole (rare)

Drug Interactions

Concurrent administration with alfentanil produce higher concentrations of alfentanil

Dose/Administration

Adult

Induction

IV: 2 - 2.5 mg/kg q 10 seconds until induction onset.

Maintenance

Infusion: 100-200 mcg/kg/min

Conscious Sedation

IV: 5 mcg/kg/min for at least 5 minutes. May increase by 5-10 mcg/kg/min every 5-10 minutes until desired level of sedation achieved

Pediatric

Induction

IV: over 3 years: 2.5 - 3.5 mg/kg over 20 - 20 seconds

Maintenance

Infusion: over 3 years: 125 - 300 mcg/kg/min

Infusion: 50-100 IU/kg every 4 hours

Special Consideration

Pregnancy Safety: B

Maintain strict aseptic technique because emulsion will support rapid growth of microorganisms.
